In this paper we will attempt at introducing the emerging concept of cell-based mechanosensation into vessel biology as the outcome of combined mechanical triggers acting in the vascular wall. In particular, we will discuss some of the pathways that translate mechanical forces into molecular signals in ECs, SMCs and vascular progenitors, and how these pathways control vascular patterning, homeostasis and, ultimately, disease.
